Leica DM750 Manual 20 Ready!.
Leica DM750 Manual 21 Substage Illumination The Leica DM750 is a vailable with two types of Substage Illumination. Identify which type of Illumination you have , as this will be important to know later . T ype 1: Standard illumination Adjustable condenser cen tering with provided tool.
Leica DM750 Manual 22 T ools used Ϙ Allen key There ar e two types of viewing tube. Identify which viewing tube you hav e: T ype 1: Leic a EZ Viewing T ube with integrated eyepieces . T ype 2: Standard viewing tube with separ ate eyepieces 1. Loosen the setscrew ( on top of the stand) using the Allen key provided .
Leica DM750 Manual 23 2. Reinstall the Leica EZ Viewing Tube ont o the stand. 3. Tighten the captiv e wingscrew using the wrench included in the delivery package.
Leica DM750 Manual 24 Leica EZ Viewing T ube – integr at ed ey epieces (con t.) Y ou can now safely r otate the L eica EZ viewing tube by loosening the captive thumbscrew , rotating the viewing tube, and tightening the captive thumbscr ew again.
Leica DM750 Manual 25 Leica Standard Vie wing T ube – separated e yepiec es 2. Secure the eyepieces in the tubes by tighte - ning the silver screws a t the bottom of the tubes using a standard Phillips screw driver (not included in the delivery pack age).
Leica DM750 Manual 26 If you wear eyeglasses f or microscope viewing, keep the rubber ey ecups folded down. If you do not wear ey eglasses, you ma y find it useful to unfold the rubber ey ecups in order to help block out ambient r oom light.
Leica DM750 Manual 27 Installing Objectives Installing Objectives When rotating the objective nosepiece , always use the knurled ring on the objective nosepiece.
Leica DM750 Manual 28 Installing the Substage Condenser Substage Condenser The Leica DM750 has an open substage condenser mount, theref ore the conden - ser needs to be installed. 1. Move the specimen stage upwar ds as far as it will go by using the coarse focusing knob on the side of the microscope .
Leica DM750 Manual 29 4. Push the substage condenser under the specimen stage into the holder by alig - ning the locating pin on the bottom of the condenser with the slot on the rear side of the fork. 5. Move the c ondenser into the highest posi - tion by using the condenser focusing knob on the left side of the stage mount.
Leica DM750 Manual 30 S et!.
Leica DM750 Manual 31 W ork Surface Always use y our microscope on a hard , stable sur face. Po wer C ord If the power cor d is not already atta - ched, attach it secur ely to the back of the microscope .
Leica DM750 Manual 32 If you have pur chased a standard L eica DM750 configuration, the condenser has already been pre-centered by L eica Microsys - tems. 1. Open the condenser aper ture by r otating the knurled ring on the condenser to the right.
Leica DM750 Manual 33 Using the Condenser The condenser is furnished with an iris diaphragm, which can be adjusted to match the effective numerical aperture of the objective 1.
Leica DM750 Manual 34 1. P osition a specimen slide on the specimen stage by sliding it under the slide grips. Slide grips hold the slide in place. 2. Using the X/Y -stage control , position the specimen slide such that a part of the speci - men is under the objective used.
Leica DM750 Manual 35 1. Rotate the objective nosepiece (using the knurled ring) in such a way that the objec - tive with the low est magnification level is rotated in to the working position. 2. Move the specimen stage upwar ds by turning the coarse focusing knob as far as it will go to the maximum position.
Leica DM750 Manual 36 Adjust the ey etubes 1. Adjust the tubes to y our interpupillary distance. F old or unfold the eyetubes to decrease or increase the distanc e between the eyepieces until y ou see one illuminated circle. The viewing tubes maintain a constant tube length for all interpupillary settings.
Leica DM750 Manual 37 If you are comf ortable wearing your corrective lenses (con tact lenses or eyeglasses) for micr oscope viewing, lea ve them on and your adjustments will be minimal.
Leica DM750 Manual 38 If your Leica DM750 is fitted with a field diaphr agm for Koehler Illu - mination, use the following pr ocedure t o assure condenser cen te - ring and focus. 1. Connect the Koehler field diaphragm to the base of the microscope so that the diaphragm leav es are in the field of view when you look through the eyepiec es.
Leica DM750 Manual 39 3. T urn the condenser centering wingscrews simultaneously to cen ter the image of the field diaphragm. 4. Open the field diaphragm until the diaphrag m leaves ar e just outside of the field of view . Koehler C onfiguration (cont .
Leica DM750 Manual 40 1. Search for the area on the specimen slide that you want t o study . 2. Move the specimen stage down t o the lowest position using the coarse adjust - ment knob. 3. Add a drop of immersion oil fr om Leica to the area of the specimen slide y ou are studying.
Leica DM750 Manual 41 Oil Immersion T echnique (cont .) 5. Slowly move the specimen stage up wards using the coarse adjustment knob until the oil drop on the slide comes int o direct contact with the lens of the oil immersion objective.
Leica DM750 Manual 42 The Leica DM750 is equipped with a time delay shutoff capability , which automati - cally turns the illumination off after 2 hours of no changes in the intensity control . Ϙ All four-position nosepiece stands ha ve the time delay shutoff enabled as a default (mostly educational applications).
Leica DM750 Manual 43 Go!.
Leica DM750 Manual 44 Now all you need to do is change objectives , set the condenser aperture (and field diaphragm if you hav e a DM750 Koehler Stand) appropriat ely for the objective magnification.
Leica DM750 Manual 45 Car e of the M icrosc ope.
Leica DM750 Manual 46 General maintenanc e Storage and handling Always use both hands to carry the microscope . There is a handle on the back of the microscope and an under cut in the front for this purpose . Ϙ The cord wr ap allows you t o wrap the cord in such a way that only the length you need is extended.
Leica DM750 Manual 47 Ϙ No part of the microscope is quite so vulne - rable to collecting dirt, dust, and oil as the front lens of the objective. Whenever you encounter lack of contr ast, cloudiness or poor definition, carefully check the condi - tion of the front lens with a magnifier .
Leica DM750 Manual 48 T r oubleshooting.
Leica DM750 Manual 49 T roubleshooting Stand The microscope does not r espond. Make sure that the socket has v oltage. Check the cable connections. Make sure that the stand is connected to the po wer supply . Check whether the fuse is defective and replace it if nec essary (see page 16).
Leica DM750 Manual 50 T roubleshooting (c ontinued) Polariza tion contrast cannot be adjusted . Bring the polarizer and analyzer int o cross position until they reach maximum darkness (without specimen). Phase contrast Phase contrast cannot be adjusted .
